AB By using quantitative RT-PCR, elevated levels of IFN gamma mRNA and
lowered IL-2 mRNA were found in PBMC of HIV-seropositive men with CD4 T
cells over 500/mm3 (mean:647/mm3) while IL-4 and IL-10 mRNAs did not
change significantly. Men whose CD4 levels fell more than 200/mm3 in the
two following years showed further changes in lymphokine mRNA levels.
Furthermore, changes in cytokine gene expression were markedly different
in CD4 and CD8 T cells from HIV-seropositive men. On a per cell basis,
mRNA levels of IFN gamma, IL-10 and TNF alpha increased approximately
three times in CD4 T cells isolated from HIV seropositive subjects (p <
0.05) while IL-2 gene expression showed no significant change. Cytokine
production in CD8 T cells, however, changed little following HIV
infection. After stimulation of PBMC with PHA, PWM, and rIL-2; IL-2 and
IL-2R were found lower, but IL-10 and TNF alpha found higher in
seropositive subjects as compared to seronegative individuals. Cytokines
play a central role in the performance and regulation of the human
immune system. The altered balance of cytokine production is likely to
contribute to the immune dysfunction in HIV-infection.
